籽瓜皮果胶的提取工艺研究

摘    要:为探究提取分离新疆籽瓜皮中果胶的最佳工艺,采用酸提醇析法探讨了原料预处理、萃取剂的种类、浓度和加入量、萃取温度、时间和浓缩比例等因素对果胶产率的影响.运用正交实验确定最佳的工艺条件为:以细渣状沸水浴灭酶3 min,硫酸为萃取剂,萃取液pH值为1.5,料液比为1:15,萃取2次,萃取温度为90℃,萃取时间分别为2.5 h和1 h,浓缩比例为1/4,冷却后经1:1.5倍乙醇沉淀,果胶产率为19.5%.

Abstract:In this paper,the best condition of extraction pectin from seed melon in Xinjiang is studied.The influences of pretrealment of raw materials,types and the concentration of extractant and adding volume,extraction temperature,extraction time and the proportion with the acid ethanol precipitation method on the yield of pectin are investigated.By orthogonal experiments the optimal conditions are obtained as follows:about 3 min in boiling water for destroying enzyme,H_2SO_4 as extractant,carring on two extractions,1.5 of pH value,1:15 of the ratio of seed melon weight to acid volume,90℃ of acid extraction temperature,2.5 h and 1 h for two extracting processes,1:4 of concentration ratio,1:1.5 of ethanol amount after cooling,then the extracting efficiency of pectin can be 19.5%.
