| 摘 要： | 基因敲除技术是研究功能基因作用的重要方法,是后基因组时代的重要研究内容.从基因敲除的细胞基础、基因敲除载体的构建及发生同源重组干细胞的筛选、基因敲除动物模型的建立、基因敲除与基因拯救、国内基因敲除技术的研究现状等方面概括了基因敲除技术的研究进展和该技术目前存在的主要问题.

| Abstract: | Gene knock out was a very important technique to study the roles of functional genes,and it was also a very important research branch in the post-genome period. In this paper,the cell basis for gene knock out, the construction of the vector for gene knock out and the screening of the embryonic stem cells where homologous recombination occured,the animal models made after gene knock out,gene knock out and gene rescue,the research progress of gene knock out at home and the problems existed were briefly introduced. |
