橄榄苦苷对高脂饮食联合链脲佐菌素诱导的糖尿病小鼠的降血糖作用研究

摘    要:目的 研究橄榄苦苷(oleuropein, OP)对高脂饮食联合链脲佐菌素(streptozotocin, STZ)诱导的糖尿病模型小鼠的降血糖作用。方法 使用高脂饮食(high-fat diet)联合STZ建立高血糖小鼠模型, 将高血糖小鼠分为模型组(HFD/STZ)及橄榄苦苷组(HFD/STZ+OP), 将正常小鼠作为对照组(chow)。对于橄榄苦苷组小鼠, 以200 mg/kg体重的剂量灌胃橄榄苦苷11周。测定三组小鼠的空腹血糖、葡萄糖耐受性以及肝脏、胰脏和附睾脂肪的质量和系数, 用H&E染色的方法检测OP对糖尿病小鼠胰岛损伤的影响。结果 橄榄苦苷能够显著降低糖尿病小鼠的空腹血糖(337.5 vs. 477.0 mg/dL); 橄榄苦苷能促进胰岛素的早期分泌, 在一定程度上改善糖尿病小鼠的葡萄糖不耐受; 橄榄苦苷增加了HFD/STZ诱导的糖尿病小鼠的胰腺胰岛面积。结论 橄榄苦苷具有良好的改善HFD/STZ诱导的2型糖尿病的作用。

Abstract:Objective Studying the hypoglycemic effect of oleuropein on high-fat diet (HFD) combined with streptozotocin (STZ)-induced diabetes model mice. Methods HFD combined with STZ was used to establish hyperglycemia mouse model, and the hyperglycemic mice were divided into model group (HFD/STZ) and oleuropein treatment group (HFD/STZ+OP). Normal mice were used as control group (chow).Oleuropein was administered by gavage at a dose of 200 mg/kg body weight for 11 weeks. The fasting blood glucose, glucose tolerance, and the weight and coefficient of liver, pancreas, and epididymal fat of the three groups of mice were measured. The effect of OP on pancreatic islet injury in diabetes mice was detected by H&E staining. Results Oleuropein could significantly reduce fasting blood glucose (337.5 vs. 477.0 mg/dL); Oleuropein can promote the early secretion of insulin, improved glucose intolerance to a certain extent; Oleuropein increased islet area in thepancreas of HFD/STZ-induced diabetic mice. Conclusion Oleuropein showed an effective improvement on type 2 diabetes induced by HFD/STZ.
