摘    要:本文采用电子显微镜对胶体金标记刀豆素与小鼠腹腔巨噬细胞表面ConA受体的结合、内吞及春在巨噬细胞中转运的形态学变化;同时采用共聚焦对巨噬细胞内Ca^2+和PH的动态变化进行了观察。

Abstract:The binding,internalization and intracellular routing of colloid gold labeled ConA(ConA Au) have been studied in mouse peritoneal macrophages by electron microscopy.The kinetic changes of cytosolic Ca 2+ and intralysosomal pH of macrophages were examined by confocal laser microscopy. Electron microscopic studies indicated that endocytosis of ConA belonged to the receptor mediated endocytosis(RME),which could be inhibited by low temperature.Endosome was the intracellular site in which the receptor and ligand uncoupled.The ligand was delivered to lysosome for degradation while the receptor recycled to the cell surface.A large number of lysosomes increased in the process of RME. Confocal laser microscopic studies indicated that cytosolic Ca 2+ increased instantanesouly and the intralysosomal pH declined and fluctuated regularly close to static value with a 10_15min interval.The findings suggested that the periodical wave of lysosomal pH is closely related to the recycling of receptors.
