Abstract:Objective:To observe the ultrastructual changes of ependymal cells in lateral recess of the fourth ventricle(LR4V) of the nitrogen monoxide deficient hypertensive rats by using SEM and TEM.Methods:30 healthy male Wistar rats were randomly divided into three groups: ① the control group(n=10) injected with normal saline;② medication 2 weeks group(n=10) and ③ medication 4 weeks group(n =10).The models of hypertensive rats were treated with nitric oxide synthase inhibitor L-nitro-L-arginine(L-NNA) with 7.5mg 2 times per day by intraperitoneal injection.Three rats were randomly selected in each group and fixed with cardiac perfusion and then the brain specimens were prepared as routine for scanning and transmission electron microscopic observation.Results: Under SEM,the surface of the ependymal cells of LR4V of control rats covered with a number of cilia,microvilli and fluid-contacting fibers,and some of the secretory granules vary in size and shape scattered in between.In 2-weeks medication group,on the surface of those ependymal cells,a special kind of "mud-like" secretions adhering to cilia can be seen.In 4-weeks medication group,clumps of "mud-like" secretions and cilia adhered together,and covered most of the cilia and the microvilli.The discharge process of "mud-like" secretions was observed as well as by TEM.Conclusions: Under electron microscope,the process of discharge of a special kind of "mud-like" secretions on the surface of the ependymal cell of LR4V was noticed.Accompanied by increase in blood pressure,the vigorous secretory activity of ependymal cells appear.It is suggested that the ependymal cells are sensitive to hypertension.
