氧化亚铁硫杆菌的菌落分离研究

摘    要:以琼脂为凝固剂 ,研究了亚铁含量、磷酸盐含量、p H值、添加硫氰酸钾对氧化亚铁硫杆菌菌落产生的影响和细菌的再培养。结果表明培养基能否有效供给酸耗用酸 ,使凝固剂不被酸降解 (胨化 ) ,使细菌能够稳定固着生长、繁殖是菌落产生的关键条件。控制好这些条件 ,可以使菌落数量比 9K固体培养基提高近两个数量级

Abstract:The effect of contents of ferrous ion and phosphate salt,pH,and potassium thiocyanate on the colony formation of Thiobacillus ferrooxidans and the subculture of the bacterrium in the presence of agar as solidifying agent have been investigated.The results have showed that the critical condition for the colony formation is whether the culture medium can steadly supply the acid for baterial oxidation to prevent agar from peptoned and keep invariable pH for the inoculum,in which the bacterium can stably anchorage on the medium for growth and multiplication;and controlling these conditions,the number of colonies can be two orders of magnitude larger than 9K solid medium.
