甘草甙的分离纯化及鉴定

摘    要:甘草甙是甘草黄酮类化合物中重要的单体活性成分,具有抗氧化、抗H IV等多种药理作用。该文在无标样条件下对甘草甙的分离纯化进行了研究,并首次采用模拟移动床色谱技术提纯甘草甙。实验表明,原料液固萃取的最佳条件为:水作萃取溶剂,原料质量浓度为40 g/L,90℃恒温水浴中萃取2.0 h。模拟移动床色谱在进样流速0.1 mL/m in;洗脱流速1.5 mL/m in;萃取流速1 mL/m in;切换时间20~21 m in;样品质量浓度0.2 g/mL;流动相V(乙醇)∶V(水)=15∶85;工作模式:1-1-2条件下(即三带模拟移动床色谱中洗脱带、精馏带、吸附带色谱柱数分别为1,1,2),可实现甘草甙的精细分离。重结晶所得产品通过质谱、紫外可见光谱、红外及核磁共振谱图对其结构进行了表征。

Abstract:Liquiritin(LQ)is one of the most important components from Glycyrrhiza flavonoids.It possesses the activities of antioxidant,anti-HIV and so on.The separation and purification of LQ are studied in this paper without standard sample.Continuous and countercurrent production can be realized by introducing simulated moving bed chromatography(SMBC) technology.The results show that the optimum conditions for raw material pretreatment are water as extraction solvent,adding 40(g/L) raw material and extracting for 2.0 h in water bath of 90 ℃.Conditions for fine separation of LQ by SMBC are as follows:feed flowrate 0.1 mL/min,desorption flowrate 1.5 mL/min,extraction flowrate 1 mL/min,switching time 20~21 min,sample concentration 0.2 g/mL and V(ethanol)∶V(water)=15∶85 as mobile phase.The column configuration in each section of three-zone SMBC are 1-1-2.The structure of recrystallization product was identified by MS,UV-vis,IR and NMR.
