替马沙星中间体7-氯-6-氟-1-(2,4-二氟苯基)-1,4-二氢-4-氧-3-喹啉羧酸合成方法的改进

摘    要:以2,4-二氯-5-氟苯甲酰基乙酸甲酸乙酯为原料,经乙氧亚甲基化、二氟苯胺胺化、环合、水解和脱羰反应对替马沙星中间体7-氯-6-氟-1-(2,4-二氟苯基)-1,4-二氢-4-氧-3-喹啉羧酸的合成方法了改进;反应条件温和,操作容易控制。并在此过程中合成了三种新的中间体化合物4,5和6。

Abstract:The research present an improved synthetic method of 7-chloro-6-fluoro-1-(2,4-difluorophenyl) -1,4-dihydro-4-oxo-quinoline-3-carboxylic acid, the important intermediate of temafloxacin. The reaction was started with 2,4-dichloro-5-fluorobenzoyl acetoformic ethyl ester, via amination, cyclization, hydrolysis and decarbonylation. The reaction temperature was moderate and the operation was easily controlled. Moreover, three new intermediates 4,5 and 6 were synthesized by the suggested method.
