盐酸羟嗪合成的研究新进展

摘    要:总结了第一代抗组胺药盐酸羟嗪及其重要中间体2-(2-氯乙氧基)乙醇和母环1-(4-氯苯基)苯甲基]-哌嗪的各种合成路线,并对各路线进行了评述,选出了一条适合工业化的路线:由1-(4-氯苯基)苯甲基]-哌嗪与2-(2-氯乙氧基)乙醇缩合,酸化,制得盐酸羟嗪.

Abstract:Various synthetic routes of the first generation antihistamines hydroxyzine dihydrochloride,its key intermediate 2-(2-chloroethoxy) ethanol and the main ring 1-(4-chlorophenyl)benzyl]-piperazine were summarized and commented.A suitable route for industrialization was selected as follows:hydroxyzine dihydrochloride was obtained through condensation of 2-(2-chloroethoxy) ethanol and 1-(4-chlorophenyl)benzyl]-piperazine,acidification.
