淫羊藿总黄酮提取物树脂纯化工艺条件的优化

摘    要:以D1400大孔树脂、HZ-841大孔树脂、PA03060聚酰胺树脂,通过动态吸附与洗脱,对淫羊藿总黄酮提取物进行纯化,HZ-841大孔树脂的纯化效果最优。通过正交试验得到最佳纯化工艺条件为:2g HZ-841大孔树脂,上样总黄酮含量2mg/mL的提取液30mL,分别以80mL水、40mL 20%乙醇、40mL85%乙醇和20mL 95%乙醇进行梯度洗脱,流速1.5mL/min。最优条件下,利用HZ-841树脂纯化淫羊藿总黄酮提取液,得到干浸膏收率3.25%,其中总黄酮含量71.8%,淫羊藿苷含量9.44%。

Optimization of Resin Purification technology of Flavonoids from Epimedium

Abstract:Contrast D1400, HZ -841, PA03060 polyamide resin for harrenwort flavonoids dynamic adsorption and dution effect, HZ - 841 is optimal. Purification conditions were obtained by orthogonal experiment as fellows :2 g HZ - 841 macro-porous resin was used to purify 30 mL solution of flavonoids content of 2mg/mL , 80mL water, dOtaL 20 % ethanol, dOtaL 85 % ethanol, 20mL 95 % ethanol gradient elution , flow rate 1.5mL/min. Under the optimized conditions, flocculation solution was purified by HZ - 841 resin,the content of total Flavonoids was 71.8% in the extract, and Icariin content was 9.44%, dry extract of Epimedium Pieces yield was 3.25%.
