溶剂萃取法分离氢化可的松和表氢化可的松

摘    要:针对工业上现有结晶分离工艺的缺点,提出了分离氢化可的松及其光学异构体表氢化可的松的新方法——溶剂萃取法。实验发现,在常规醇类、酮类、酯类、醚类、氯代烃类和芳香烃等溶剂中,乙酸丁酯具有较好的萃取分离性能,以乙酸丁酯为萃取剂用溶剂萃取法分离氢化可的松粗品可获得纯度为98%以上的氢化可的松,已达到工业生产水平。文中还研究了乙醇含量、温度、盐析效应和pH对分配的影响。

Abstract:Solvent extraction as a new method for separation of hydrocortisone and its optical isomer,epi hydrocortisone,is suggested instead of traditional crystallization operation.The extraction behavior of alcohols,ketones,esters,ethers and chloro carbons was studied experimentally.It is found that the better solvent is butyl acetate for industrial separation process.The distribution coefficients of hydrocortisone and epi hydrocortisone are 10.0 and 2.7 respectively in butyl acetate/water system at 17℃.When butyl acetate solution of the mixture of hydrocortisone,epi hydrocortisone and other steroid impurities was scrubbed by deionized water in 9 stage cross current,the purity of hydrocortisone in organic phase increases from 78%~98.2%(wt) and attained the standard of medical cortisol.Effects of alcohol concentration,temperature,salting out and pH on extraction were also discussed.
