摘    要:以海藻酸钠为载体,还研究了固定化细胞的制备,对接种环数、种子培养时间进行了优化,还研究了不同乙醇初始浓度对醋酸产量、菌体个数、菌体生长周期、葡萄糖消耗量、乙醇消耗量的影响。获到如下结论:优化接种数为15环、培养时间为36~48h之间;葡萄糖浓度与OD值的换算公式:Y=4.723 7x;菌体个数与菌液OD值的相关性:y=402.88x;体积分数5%乙醇时发酵液醋酸转化率最高,最佳培养时间为96h;乙醇体积分数越高,菌体生长延迟期越长;乙醇的含量越高,则葡萄糖的消耗就越少;培养时间为72h时,乙醇消耗完毕,由于中间产物乙醛的存在,醋酸在72~96h时间段继续生成。

Abstract:As the vector of sodium alginate, this paper studys the preparation of immobilized cell, optimizes the number of inoculation rings and culture time, and also researches the effect of the yield of acetic acid, the mumber of bacteria cell, the cell growth rate, the glucose consumption, and the alcohol consumption under the different initial concentration of alcohol. We obtain the conclusions as follows: the best number of inoculation is 15 rings, and culture time is between 36~48 h; the conversion formula of the concentration of glucose and OD value: y=4. 723 7x; the conversion formula of the number of bacteria cells and OD value of bacterial liquid: y=402.88x; the 5 % alcohol concentration of acetic acid fermentation broth produces the highest conversion rate, and the best time to culture is 96 h; as the alcohol concentration is higher, the cell growth plays an extended period of growth delay; the higher the alcohol content is, the smaller the glucose consumption is; after fermenting 72 h, alcohol consumption has finished, but because of the existence of acetaldehyde the intermediate, acetic acid in 72~96 h continues to generate.
