工业生产微生物谷氨酰胺转胺酶的分离工艺研究

摘    要:研究以Streptoverticillium mobaraense菌株发酵产生的谷氨酰胺转胺酶的分离工艺.通过发酵液分离处理后的收率和比酶活测定,确定超滤和乙醇沉淀的最适条件.试验结果表明:超滤的最适浓缩倍数为2.5~3.2;浓缩液pH值为8,冷乙醇按照酶与乙醇体积比1:1.5加入为最佳分离条件.

Abstract:The separation technics of transglutaminase produced by Streptoverticillium mobaraense was analyzed. By comparing the enzyme activities and the enzyme yields in different conditions, the optimum condition of ultrafiltration and deposition with organic solvent was confirmed. The results showed: the best condition of the enzyme separation with organic solvent is that the ultrafiltrate was concentrated 2.5 to 3.2 times, then adjust the pH to 8, and add 1.5V refrigerant ethanol.
