摘    要:用薄膜冷冻法制备了克拉霉素脂质体,以高效液相色谱法为分析手段,色谱柱采用Hyper-sil ODS柱(250 mm×4.6 mm,5μm);流动相为乙腈与0.1 mol/L磷酸盐缓冲液(体积比为70∶30),用稀磷酸调节pH值至4.8,流速1.0 m/min;检测波长205 nm.克拉霉素在10-200 mg/L内浓度与峰面积呈良好线性关系,相关系数r=0.999 7.采用反透析法测定脂质体的包封率,并确定了透析平衡时间,讨论了水合介质和冻融次数对包封率的影响.

PREPARATION OF SOYBEAN LECITHIN CLARITHROMYCIN LIPOSOME BY THIN FILM AND FREEZE-THAWING METHOD

Abstract:The clarithromycin liposome was prepared using the thin film and freeze-thawing method and was analyzed with HPLC.HPLC analysis was performed on a Hypersil ODS column(250mm×4.6mm,5μm particle size),using acetonitrile and 0.1mol/L the phosphate buffer(pH4.8)(70∶30,V/V) as the mobile phase.The flow rate was 1.0mL/min.The absorbance of the eluate was monitored continuously at 205nm with a PDA.The line arrange of clarithromycin was 10~200mg/L(R=0.9997).The entrapment efficiency of clarithromycin liposome was determined by reverse dialysis method,and the dialysis equilibrium time was determined.The effect of the hydration medium and the freezing and thawing times on entrapment efficiency was discussed.
