离子交换纤维纯化银杏叶黄酮影响因素的正交法研究

摘    要:采用正交法考察了各因素对浸取纯化黄酮工艺的影响,确立了离子交换纤维纯化黄酮的最佳工艺.获得浸取银杏叶黄酮的最佳条件:以pH值为8的70%的乙醇溶液,在75℃浸提3.5h;强碱阴离子交换纤维吸附黄酮的最佳条件:上柱药液的pH=4,药液流速0.38mL/min,药液质量浓度0.5289mg/mL,固液比1:200;吸附在强碱阴离子交换纤维上的黄酮动态解吸的最佳条件:HCL浓度2mol/L,流速0.5mL/min,解吸剂是体积分数为60%的乙醇,体积60mL.实验结果表明用强碱性阴离子交换纤维提纯黄酮方法可行.

Abstract:To optimize the purification process of flavones, the effects of factors on purification of ginkgo were studied by orthogonal test. The results showed that the optimal conditions of extracting flavones were as the following: the pH value is 8, the solvent is ethanol of 70 percent, the temperature is 75 ℃ , and the time is 3.5 hours . The optimal conditions of dynamic adsorption were got as the following: the pH value is 4, the velocity is 0.38 mL/min, the mass concentration of flavones is 0. 528 9 mg/mL and the ratio of solid to liquid is 1 : 200. The .best dynamic desorption process parameters is that the concentration of HC1 is 2 mol/L, the velocity is 0.5 mL/min, the desorption agent is ethanol with the volume of 60 mL and the volume fraction was 60%. The results indicated that the purification of flavones using ion exchange fiber was feasible.
