摘    要:采用单因素法,以MRS培养基为基础培养基,对短乳杆菌P-347产细菌素的适宜碳源和氮源的种类、氮源的浓度、起始pH和培养温度进行了初步探索。以金黄色葡萄球菌AS1.72为指示菌,采用牛津杯定量扩散法检测发酵上清液的抑菌活性。结果表明,短乳杆菌P-347以葡萄糖为碳源,以大豆蛋白胨为氮源,在起始pH 5.0、37℃培养24h,可获得最大菌体密度;以2%葡萄糖为碳源,以1%酪蛋白胨为氮源,在起始pH 6.5、30~37℃培养24h,发酵上清液的抑菌活性最大,抑菌圈直径为32.5mm。

Abstract:The influence of carbon and nitrogen sources,nitrogen concentration,initial pH and temperature on cell growth and bacteriocin production of LactobacillusbrevisP-347 was investigated by single factor test.The antimicrobial activity of bacteriocin was detected against Staphylococcus aureus AS1.72 by means of the oxford plate assay system.The highest cell density could be obtained when Lactobacillus brevis P-347 was cultured in glucose and soy peptone for 24 hat 30 ℃ and initial pH 5.0.The highest antimicrobial activity in culture supernatant was obtained at 30-37 ℃ and initial pH 6.5for 24 hwith 2% glucose as carbon source and 1%casein peptone as nitrogen source,in which the inhibitory zone is 32.5mm diameter.
