应用于固定化生物技术的高效脱酚菌的分离和鉴定

摘    要:目的为了获得处理煤气厂含酚废水的高效菌株,解决含酚废水中总酚降解问题.方法从哈尔滨气化厂现有生化处理系统的曝气池活性污泥中筛选高效脱酚菌.进行生理生化鉴定和16SDNA测序。建立了系统发育树.对高效脱酚菌进行了多组分苯酚羟化酶大亚基因(LmPHs)的扩增,采用生物固定化技术将高效脱酚菌固定在活性炭上,形成固定化生物活性炭(IBAC)。应用于哈尔滨气化厂煤气废水处理的中试设备中.结果从活性污泥中筛选出3株高效脱酚菌,其降解废水中总酚的能力均在99%以上。IBAC段进水COD的质量分数在600mg/L以下,总酚的质量分数在75mg/L以下时,IBAC段对COD和总酚的去除率可以分别达到80%和70%以上.鉴定出这3株菌分别属于气单胞菌属(Aeromonas)、芽孢杆菌属(Bacillus)和不动细菌属(Acinetobacter).结论对3株高效脱酚菌的继续研究可使其在含酚的污水处理等实际运用中起到重要的作用.

Abstract:The efforts to work on dominant bacteria and remove the phenolic wastewater were carried out in the study.The strains of phenol degradation bacteria with high efficiency were screened from activated sludge of the aeration basin of the present biochemical treatment system in Harbin Gasification Plant by advanced bio-engineering technology.With the aid of sequencing the 16SDNA and establishing the phylogenic tree,Based on screening and acclimating phenol degradation strains,LmPHs fractions of the highly-effective strains of phenol degradation bacteria were amplified.The strains were fastened to biological activated carbon(IBAC) by biological immobilization technologies.They were already applied in the medium tested equipments of processing coal-gas wastewater in Harbin Gasification Plant.3 strains of phenol degradation bacteria with high efficiency were screened by media from sludge and the phenol removal rates were higher than 99%.When influent COD of IBAC reactor was below 600?mg/L and phenols of IBAC reactor below 75?mg/L,COD and phenol removal rates were higher than 80% and 70%.They were identified physiologically and biochemically.They were respectively Aeromonas,Bacillus and Acinetobacter.So it is sure to happen that the subsequent research will make 3 strains play an important protective role in the environment.
