酶注射式葡萄糖生物传感器的研制

摘    要:葡萄糖是发酵过程中菌体生长和产物合成的主要碳源。为了稳定产物产量与质量,必须控制葡萄糖在发酵液中的浓度。为了解决现有"固定化酶式"葡萄糖生物传感器难以用于发酵过程葡萄糖在线测量的问题,提出了"酶液式"葡萄糖生物传感器,研制了适用于发酵过程的、在线使用的酶注射式葡萄糖生物传感器。实验结果表明:传感器线性范围为0~100mg/dL,且线性相关系数R2为0.9990,传感器灵敏度为2.59nA/(mg/dL),传感器检测限为0.07mg/dL;传感器重复性较好,对50mg/dL葡萄糖浓度,变异系数(CV)为2.02%。所研制传感器基本满足了发酵过程葡萄糖在线测量的要求。

Abstract:Glucose is the main carbon source of bacteria growth and product synthesis in fermentation process.In order to stabilize the product yield and quality,glucose concentration in the fermentation broth must be controlled. At present,it is difficult to realize glucose on-line measurement by enzyme-immobilized glucose biosensor.In order to solve this problem,enzyme-liquid glucose biosensor is proposed.Enzyme-injected glucose biosensor,which is used for fermentation process glucose on-line measurement,is developed.Experimental results show the linear range of the sensor is 0~ 100 mg / dL,a correlation coefficient R 2 is 0.999 0,the sensitivity is 2.59 nA /(mg / dL), the detection limit is 0.07 mg / dL and the coefficient of variation is 2.02 % at the glucose concentration of 50 mg / dL.The sensor basically meets the requirement for glucose on-line measurement in the fermentation process.
