哈茨木霉菌Tri101基因的克隆与序列分析

摘    要:根据拟枝孢镰刀菌等真菌中Tri101的保守序列设计简并引物,利用简并PCR和基因组步行法克隆了哈茨木霉菌Tri101基因.结果表明:Tri101基因的开放阅读框为1 314 bp,无内含子存在,与预计的Tri101序列特征一致.该基因编码蛋白质由437个氨基酸组成,预测相对分子质量约为4.75 kD,系统进化树显示其与棒曲霉(Aspergillus clavatus)NRRL 1亲缘关系最近,与NRRL 1中3-O-乙酰转移酶的的蛋白质同源性达到55%.Blast数据库分析表明其属于转移酶总科,单端孢酶烯乙酰转移化酶.以禾谷镰刀菌(Fusarium graminearum)TRI101蛋白(Parent PDB:3b2s Chain:A)的晶体为模板进行了同源建模,为进一步研究该基因的功能奠定基础.

Abstract:Trichoderma harzianum Tri101 gene encodes a trichothecenes 3-O-acetyltransferase that converts trichothecenes to less toxic form by transferring acetyl.Trichoderma harzianum Tri101 was cloned by using degenerate PCR and Genome walking approach.Results showed that the ORF of Tri101 was 1 314 bp in size and encoded 437 amino acids.No intron was detected in sequence as we expected.The molecular weight of the putative Tri101 was 4.75 kD.The result analysis by Blast showed that it belonged to transferase superfa...
