牙鲆选择性养殖群体遗传结构的微卫星分析

摘    要:应用10对微卫星引物对三个牙鲆群体各30个个体进行了群体遗传多样性分析.普通群体的平均等位基因数为7.6,显著高于感病群体的5.80,略高于抗病群体的7.2.普通群体的10个位点的平均基因型数为11.4,而感病群体和抗病群体分别只有8.3和9.0.普通群体的平均有效等位基因数为3.97,略高于感病群体和抗病群体的3.78和3.72.普通群体10个位点最高频率等位基因的平均频率值为0.388,而感病群体和抗病群体分别为0.407和0.425.普通群体低频等位基因(0-0.05)总共有27个,而感病群体和抗病群体分别只有15个和19个.普通群体共得到14个私有等位基因,感病群体和抗病群体仅各得到3个等位基因.研究结果表明了感病群体和抗病群体两个选择性养殖群体遗传多样性的降低.

Abstract:Ten microsatellite loci were used to analyze one control population and two selected cultured populations of flounder Paralichthys olivaceus. A total of 76 different alleles were obtained over all loci. The number of alleles per locus ranged from 3.67 to 8.67. The average of observed and expected heterozygosity ranged from 0.610 to 1.000, and from 0.536 to 0. 821, respectively. A total of 20 unique alleles were found in all the three populations. The effective number of alleles varied from 2.14 for Po35 to 5.60 for Po91. The number of genotypes ranged from 5.33 for Po56 to 12.33 for Pol. Compared with the control population, the two selected cultured populations, susceptible and resistant, showed significant genetic changes such as a fewer alleles per locus, a smaller number of low frequency alleles, a smaller number of unique alleles and a smaller number of genotypes, which indicated a reduction in genetic diversity.
