| 摘 要： | 分别将IL-6和HIV-1gag基因插入到鸡痘病毒表达载体质粒pUTAL串联启动子和复合启动子(ATI-P7．5)下游，构建了重组鸡痘病毒表达质粒pUTA-GAG-IL6。经同源重组和Western blot检测，获得重组鸡痘病毒，并将该重组病毒免疫BALB／c小鼠，检测免疫小鼠血清抗体水平和特异性CTL杀伤活性。结果获得的重组鸡痘病毒可同时表达HIV-1核心蛋白和IL-6，并在感染细胞内形成病毒样粒子。重组病毒具有良好的免疫原性，可诱导小鼠产生特异性体液免疫和细胞免疫。

| Abstract: | To construct the recombinant fowlpox virus expression plasmid pUTA-GAG-IL6, IL-6 and HIV-1 gag gene were inserted into the downstream of P7.5 tandem promoter and combined promoter ATI-P7.5 of fowlpox virus expression vector pUTAL respectively. The recombinant fowlpox virus (rFPV) was obtained by homologous recombination and Western blot analysis,and the level of serum antibody in mice and the cytotoxic activity of specific CTL were examined after the mice were immunized with rFPV. The results showed that rFPV could coexpress HFV-1 Gag protein and IL-6,and VLPs were formed in the cells infected by rFPV. rFPV possessed good immunogenicity and could elicited humoral and cellular immunity in mice. |
