大鼠急性铀中毒及螯合剂治疗后肾脏损害的透射电镜与扫描电镜观察

本文报道了利用透射电镜与扫描电镜观察大鼠急性铀中毒及螫合剂治疗后的肾脏损害。急性铀中毒以后可以出现肾近曲管上皮细胞线粒体肿胀或浓缩、细胞基质水肿、细胞间隙扩张、基底褶消失、底网增厚、基膜密度加大、肾小球足细胞足突肿胀、毛细血管内皮孔异常等一系列与细胞内液体转输障碍有关的超微结构变化。单纯使用螯合剂 CaNa_3-DTPA 或 H-73-10可以引起细胞内液体潴留或溶酶体内沉积致密颗粒。铀中毒极期使用这两种螯合剂治疗,对防止铀中毒的肾坏死不但无益相反有害;这可能是由于它们加重了细胞内液体转输系统和溶酶体的负担。

TEM AND SEM OBSERVATION OF URANIUM INDUCED RENAL NECROSIS AND THE RESULT OF CHELATES TREATMENT ON RATS

Ultrastructural changes in kidney related with the impairm-ent of intracellular fluid transportation can be found after acute uraniumintoxication in rats,such as:condensation and swelling of mitochondria,matrix edema,dilatation of intercellular space,disappearance of basalfolds,thickening of basal web,intensification of basal lamina of theproximal convoluted tubule epithelium cells,and foot processes swelling,diminishing of endothelium fenestrae of the renal glomerulus.Heavy metalchelates DTPA and H-73-10 treatment may result in intracellular fluidaccumulation and condensed grannule formation in lysosome.Treatmentwith these chelates in the critical stage of uranium intoxication mayaccelerate the necrosis instead of diminishing.This may be related to theaugment of the load of lysosome and intracellular system of fluid transpor-tation.
