Abstract:In order to make full use of Ganoderma lucidum and increase the yield of polysaccharides,carboxymethylation of polysaccharides was carried out.Bailey powder machine was applied to make ultrafine Ganoderma lucidum powder(UGLP).Carboxymethylation was progressed in ethanol-water medium by one-step method and the degree of substitution(DS) was determined by copper salt precipitation method.The structure of carboxymethyl Ganoderma lucidum polysaccharides(CGLP) was identified by FTIR.And the optimal conditions were: ethanol concentration 80%,2.5 mol/L concentration of NaOH,1.55 mol/L concentration of monochloroacetic acid(MAC),reaction temperature 55 ℃,reactio n time 4 h,and the DS was 0.81 under these conditions.In addition,water-solubility of CGLP was proportional to the carboxymethyl DS and the yield of water-soluble polysaccharides was about 40%.Tthe one-step carboxymethylation of GLP was an effective method to enhance the yield of polysaccharides and their water-solubility.
