Abstract:A method was developed for the determination of Para Red and Sudan dyes in food by reversed phase-high performance liquid chromatography(RP-HPLC) with a new simple extract system.Sample was extracted from food with methanol-acetonitrile-acetone.The analysis was performed on a Shiseido C18 column(150 mm×4.6mm i.d.,5μm),with water-acetonitrile as the mobile phase at a flow rate of 1.0mL/min.Detection was carried out at the wavelength of 500nm and confirmed by spectra.All of the six compounds demonstrated good linear relationship(r=0.9998).The limit of detection(LOD)(spiked at the levels of 0.5~5.0 μg/mL) was 0.1 mg/kg.The average recoveries for all six dyes were ranged from 85%~106%,and the relative standard deviations were form 2.14%~4.82%.The method is sensitive,reliable and can be applied for the analysis of Para Red and Sudan dyes in food.
