藤茶水溶性多糖的分离纯化及性质的研究

摘    要:经水提、醇沉、Sevag法脱蛋白的藤茶多糖(AGP),通过DEAE-SephadexA-25和SephadexG-200柱层析,得到多糖AGP-3和AGP-4。HPLC和SephadexG-200柱层析鉴定AGP-3和AGP-4为分子量相对均一的蛋白多糖,硫酸-咔唑反应、考马斯亮蓝G-250反应均呈阳性。

Abstract:Polysaccharide from Ampelopsis grossedentata (AGP) was obtained after being extracted with hot water, precipitated with alcohol and removed free protein with Sevag method. AGP was isolated and purified by DEAE-Sephadex A-25 and Sephadex G-200 column chromatography, two pure polysaccharides (AGP-3 and AGP-4) were gained. The results of HPLC and Sephadex G-200 column chromatography illustrated that both AGP-3 and AGP-4 were protein-bond polysaccharids with comparatively homogenous molecular weights. Also, the two polysaccharids were colored by coomassie brilliant blue G-250 and carbazole reaction.
