摘    要:为进一步提高环脂肽的产量,应用单因素试验与正交设计对Bacillu ssubtilis0w-097产环脂肽深层液体发酵培养基组成及发酵条件进行了优化。优化的发酵培养基组成为:玉米粉4.0%、豆粕粉3.0%、CaCl21.2g/L、KH2PO42.0g/L、Na2HPO48.0g/L和起始pH值为8.5。最佳培养备件:装瓶量为125mL,菌龄为24h,接种量为4%,培养温度为37℃,摇床转速为200rpm。在此优化条件下环脂肽的产量为1.026g/L。

Abstract:In order to enhance the production of cyclic lipopeptide, its fermentation conditions are studied. With single factor experiment (SFE) and orthogonal design experiment ( ODE ), the medium composition of CLP - producing strain - - Bacillus subtilis Ow - 097 in the submerged liquid fermentation and its fermentation conditions were examined. The final results indicated the optimized medium compositions as the following : corn meal 4.0% , soybean cake powder 3.0% ,CaCl2 1.2 g/L,KH2PO4 2.0 g/L, Na2HPO4 8.0 g/L, and initial pH 8.5. The optimal culture conditions were set as the following: media amount 125 mL, cell age 24 h, inoculation 4%, culture temperature 37 ℃, and rotation speed 200 rpm. Under the above conditions,the production of cyclic lipopeptide is 1. 026g/L.
