姜油树脂各部位体外抗氧化活性的比较

摘    要:为了比较姜油树脂及其大孔树脂分离纯化的各部位体外抗氧化作用,利用70%乙醇溶液浸提生姜得到姜油树脂,再用AB-8大孔树脂对其分离纯化并用香草醛作为标准品测定各部位姜辣素含量,利用Fe2+-H2O2诱导小鼠肝匀浆生成MDA,测定样品对MDA的抑制率,并与VitE进行比较,模拟机体黄嘌呤-黄嘌呤氧化酶反应系统测定样品类SOD活性。结果显示:50%乙醇相姜辣素含量最高,为34.7%;姜油树脂和50%相、75%相、95%相对MDA均有不同程度的抑制,50%相和75%相效果优于VitE,水相和25%相在实验浓度范围内达不到半数抑制浓度;在类SOD测定中,50%相和75%相类SOD活性最强,二者无显著差异,其它各部位类SOD活性偏低。因此,姜油树脂和各部位体外有一定的抗氧化活性,尤以50%相和75%相最为明显,且存在剂量效应关系。

Abstract:Objective:To compare the antioxidant activity of ginger oleoresin and different parts separated by macroporous resin.Methods:first obtain the ginger oleoresin from 70% ethanol solution and then examine the gingerol content of each part with vanillin as the standard after being separated and purified by AB-8 macroporous resin;Using Fe2+-H2O2 induced liver homogenate to generate MDA;Determining the inhibition rate of the samples on MDA,and comparing it with vitamin E;Assaying the similar SOD activity by simulating body's xanthine-xanthine oxidase system.Results:Fifty percent ethanol region contains the highest gingerol,which is 34.7%;Ginger oleoresin and other parts except for the extractive by water and 25% ethanol had different inhibition rates;compared with Vitamin E,50% ethanol region and 75% ethanol region were more effective;water region and 25% ethanol region were not effective in the experimental concentration range.In the determination of the similar SOD activity,the similar SOD activities of 50% ethanol region and 75% ethanol region were the best,without showing any significant difference.Conclusion: All the samples have antioxidant activity with a Dose-response relationship,especially the 50% ethanol region and 75% ethanol region.
