葛根抗氧化活性有效部位的筛选研究

摘    要:对葛根提取物及其萃取物进行活性成分含量测定及抗氧化活性和总还原能力的测定。以DPPH自由基清除能力、羟基自由基清除能力和总还原能力代表其抗氧化活性,比较葛根各提取物及萃取物的抗氧化活性。采用Origin软件中的多项拟合计算活性试验的IC50值和A0.7值,并采用Pearson相关性分析测定活性成分含量与抗氧化活性相关性。最终测得结果表明,葛根70%乙醇提取物中含量最高的是总黄酮,含量为326.1 mg/g,水提物中总多糖含量最高,为183.9 mg/g。葛根提取物及其萃取物抗氧化活性顺序为:乙酸乙酯相>正丁醇相>水相。葛根70%乙醇提取物的乙酸乙酯萃取物的抗氧化活性最优。

Abstract:The active ingredient content,antioxidant activity and reducing ability of Pueraria lobate extract and its extract were determined.The antioxidant activity was represented by DPPH radical scavenging ability,hydroxyl radical scavenging ability and reducing ability,was used to compare the antioxidant activity of various extracts and extracts of Pueraria lobate.The IC50 value and A0.7 value of the activity experiment were calculated by multiple fittings in Origin software,Pearson correlation analysis was used to determine the correlation between the content of active ingredients and antioxidant activity.The final measurement results showed that the highest content in the 70%ethanol extract of Pueraria lobata was the total flavonoids,the content was 326.1 mg/g,and the highest total polysaccharide content in the water extract was 183.9 mg/g.The order of anti-oxidation activity of Pueraria lobata extract and its extract was:ethyl acetate phase>n-butanol phase>water phase.Ethyl acetate extract from Pueraria lobata 70%ethanol extract had the best antioxidant activity.
