烟草上马铃薯Y病毒的RT-PCR检测

摘    要:本文根据已报道的PVY CP基因序列,设计合成了2套扩增PVY CP基因全序列和中间部分序列的寡核苷酸引物,用两套程序分别对马铃薯Y病毒普通株系(PV YO)和马铃薯Y病毒坏死株系PV YN) CP基因进行RT-PCR扩增,结果分别得到了与预期大小一致的804和301bp片段,健康植株对照中未扩增到任何产物,建立了RT-PCR检测烟草上马铃薯Y病毒的程序。

Abstract:Two paris of primers were designed and synthesized based on the nucleotide sequence of the CP gene of potato virus Y (PVY) genome. The CP gene of PVY+O and PVY+N were amplified by RT-PCR according to two differential procedures respectively. The target fragments about 804 and 301 bp were amplified from the infected samples, but not from health sample. The method of detecting PVY affecting tobacco was built.
