南方根结线虫侵染烟草抗感品种前后的代谢组学分析

摘    要:为明确南方根结线虫侵染烟草抗感品种前后代谢途径的差异,选取烟草抗病品种G28与感病品种长脖黄壮苗为试验材料,设置4个处理:1)G28接种J2s悬浮液(G_RKN);2)长脖黄接种J2s悬浮液(C_RKN);3)G28接种清水(G_CK);4)长脖黄接种清水(C_CK)。每个处理3次重复。采用LC/MS技术分析接种前后代谢通路的变化。结果表明,南方根结线虫侵染的抗、感病品种G28与长脖黄在抗病相关的代谢通路都有明显变化,鉴定出的差异代谢物包括了生物碱、脂肪酸、类黄酮、萜类和聚酮等物质。其中抗病品种被显著激活的代谢通路有10条,而感病品种只有5条。所以抗、感品种被南方根结线虫侵染后防御能力不同。

Metabonomics analysis of resistant suceptible tobacco varieties before and after infection by Meloidogyne incognita

Abstract:To clarify the difference of metabolic pathways of resistant tobacco varieties before and after infection by Meloidogyne incognita, experiments were conducted using the resistant tobacco variety G28 and the susceptible variety Changbohuang seedlings as test materials. A total of 4 treatments were set up: 1) G28 inoculated with J2S suspension (G_RKN); 2) Changbohuang inoculated with J2s suspension (C_RKN); G28 inoculated with water (G_CK); 4) Changbohuang inoculated with water (C_CK). Each treatment was repeated 3 times. The changes of metabolic pathways before and after inoculation were analyzed by LC/MS technique. It was confrmed that the disease resistance-related metabolic pathways of resistant varieties G28 and susceptible varieties Changbohuang infected by Meloidogyne incognita were signifcantly changed, and the differential metabolites including many alkaloids, fatty acid, flavonoids, terpenoids, polyketones and other disease-resistant related substances had been identifed. Among them, 10 metabolic pathways were found signifcantly activated in the resistant cultivar, and only 5 of them in the susceptible varieties. From the perspective of metabolome, the difference in defense ability of resistant varieties after infection was revealed.
