摘    要:对选育的高产曲酸菌(编号Co-26)进行分离和分子鉴定。本实验采用分离高产曲酸菌株(编号Co.26),获得纯化的菌丝体,利用真菌通用引物ITS1和IITS4扩增菌株rDNA的ITS区,扩增产物并进行测序,测序结果在GenBank中进行同源性搜索,选取部分具有代表性的ITS序列,利用软件MEGA5.1构建系统发育树,通过序列比对分析得出高产曲酸菌为米曲霉。

Abstract:The separation and the molecular identification of the high yield Kojic acid bacteria (Numbers Co-26). This experiment got the punhed mycelia by separating the high yield Kojic acid bacteria (Numbers Co-26), and then got this strains's rDNA ITS area DNA sequence by using the common primer ITS1 and general ITS4 for fungi to amplify this area, sequencing the PCR production and finally blasting the homology sequence in GenBank in homology search. With the selected part of ITS typical sequence, and the software MEGA5.1 , we built the phylogenetic tree of this Ko- jic acid bacteria, through the sequence alignment analysis and thus got the high yield Kojic acid bacteria: Aspergillus oryae.
