摘    要:本文研究了中性蛋白酶基因在不同宿主菌及各种培养条件下的蛋白酶表达水平。用含有中性蛋白酶基因的重组质粒PMPR4,PMPR8和PMPR16转化BacillussubtilisBG2036,BacillussubtilisAS1.398和BacillussubtilisML,对转化菌产生的蛋白酶活性分析表明:三个重组质粒的导入,均使主菌的酶活性有大幅度的提高。其规律是:宿主菌的蛋白酶活性越低,提高的幅

Abstract:This paper presented the study of expression of neutral protease gene from Bacillus Subtilis in various host cells and different culture conditions. The isolated plasmid PMPR4. pMPR8 pMPR16. harboring the neutral protease gene were used to transform four B. subtilis strains ASl. 398, ML,DB104 and BG 2036. The protease activity of transformatnts showed a significant increase. And the lower the protease activity of the host cell was, the larger the increasing scale was, the higher protease activity of the host cell had, the higher the protease activity of transformant was obtained. The transformants exhibited a various protease activity when they were incubated in different conditions,such as in different pH, temperature. growing time, antibiotic as well as the antibiotic oconcentration, etc. The results implied that the different antibiotic and their concentration had an obvious effect on the protease gene expression; the incubation temperature and time also effected strongly the expression of protease gene.
