摘    要:为研究菌株Photobacterium sp. LG-1理化性质,提高其产酶活性。利用BIOLOG对其进行生理生化实验,通过紫外诱变、化学诱变及复合诱变进行育种,并对其进行酶学性质及抑菌性研究。结果表明,该菌株复合诱变后酶活达2.690 U/mL,酶活提高率为139.78%;最适反应温度为35 ℃,0 ℃仍具有催化活性;最适反应pH为8.0,在偏碱性环境中有较好的稳定性;Ca2+、K+、Mg2+、Cu2+、Na+对几丁质酶活性有促进作用。LG-1对根霉、黑曲霉、毛霉及玉米腐烂病有抑制活性。

Abstract:To study the physicochemical properties of the strain Photobacterium sp. LG-1 and improve its enzyme production activity, BIOLOG was used to carry out physiological and biochemical experiments. Parallel breeding through ultraviolet mutagenesis, chemical mutagenesis and compound mutagenesis. Research on its enzymatic properties and antibacterial properties. The enzyme activity of this strain reached 2.690 U/mL after compound mutagenesis, and the increasing rate of enzyme activity was 139.78%. The optimum temperature was 35 °C, and the chitinase still showed catalytic activity at 0 °C. The optimum pH was 8.0, and it had stability in an alkaline environment. Ca2+, K+, Mg2+, Cu2+ and Na+ could promote chitinase activity. LG-1 had inhibitory activity against Rhizopus, Aspergillus niger, Mucor and rot of Corn .
