复合诱变灵芝原生质体选育高产胞外多糖菌株

摘    要:分别采用紫外线(UV)+甲基磺酸乙酯(EMS)、UV+氯化锂复合诱变灵芝原生质体,对高产胞外多糖的菌株进行了筛选。结果表明:UV+EMS最佳诱变条件为UV照射10s,EMS质量分数为0.06%,胞外多糖产量最高为2.95mg/mL,较原发菌株提高了49.75%;UV+氯化锂最佳诱变条件为UV照射20s,氯化锂质量分数为0.02%,胞外多糖产量最高为3.11mg/mL,较原发菌株提高了57.87%;2种诱变方式中,最佳诱变条件下,UV+氯化锂复合诱变所获得的胞外多糖产量较UV+EMS复合诱变的高5.42%。诱变株FE3第3、5代胞外多糖产量较原发菌株分别提高了40.10%、25.38%,诱变株FL10第3、5代胞外多糖产量较原发菌株分别提高了51.78%、30.96%,都具有较好的遗传稳定性。

Abstract:Protoplasts of Ganoderma lucidum were treated by compound mutation,ultraviolet radiation(UV) and ethylmethane sulfonate(EMS),ultraviolet radiation and lithium chloride.Mutation strains with higher polysaccharide yield were screened.The results showed that the optimum mutation condition of UV and EMS was radiation for 10 s and 0.06% EMS with the highest yield of polysaccharide,2.95 mg/mL,and 49.75% higher than original strain.The optimum mutation condition of UV and lithium chloride was radiation for 20 s an...
