姜黄素-壳聚糖-核酸体系的荧光增强效应及其分析应用

摘    要:研究了以壳聚糖(CTS)-姜黄素(CU)为探针测定脱氧核糖核酸(DNA)的新方法。研究表明,在甘氨酸-HCl缓冲溶液中,pH2.90的条件下,在CTS存在下,脱氧核糖核酸能增强姜黄素的荧光,并且其荧光增强程度与脱氧核糖核酸的浓度在一定范围内呈良好的线性关系。在最佳实验条件下,鲱鱼精子脱氧核糖核酸hsDNA和小牛胸腺脱氧核糖核酸ctDNA的检出限分别达到7.20×10-9g/mL和3.60×10-9g/mL。同时采用共振光散射、吸收光谱等方法对其作用机理进行了探讨。

Abstract:It is proposed that chitosan(CTS)-curcumin(CU) may be a new fluorimetric probe to determine the concentration of DNA.Experiments indicate that in the pH2.90 Glycine-HCl buffer solution,the fluorescence intensity of curcumin(CU) can be highly enhanced by DNA in the presence of chitosan(CTS),and the enhanced intensities of fluorescence are quantitatively in proportion to the concentrations of DNA in a certain range.Under the optimum conditions,the detection limits(S/N=3) are 7.20×10-9 g/mL and 3.60×10-9 g/mL.Resonance light scattering,ultraviolet and other means are used to discuss its mechanism.
