硫酸化鸡油菌多糖的体外抗氧化作用研究

摘    要:用超声波法提取鸡油菌多糖CCP,经浓硫酸法对其进行硫酸化修饰,得到硫酸酯鸡油菌多糖S-CCP。通过测定对超氧阴离子、羟自由基的清除率,以及对H2O2诱发MDA生成的抑制和H2O2诱发红细胞溶血的影响,评价S-CCP的体外抗氧化作用。实验结果表明,S-CCP较之CCP具有明显的体外抗氧化作用。当多糖浓度为5mg/mL时,S-CCP对O2·清除率高出CCP22.71%;多糖浓度0.4mg/mL时,S-CCP对·OH清除率高出CCP14.9%;多糖浓度4mg/mL时,S-CCP作用于H2O2诱发MDA的生成量低于CCP0.4nmol/mg,且其对H2O2诱发红细胞氧化溶血抑制率高出CCP33.9%。

Abstract:Polysaccharide (CCP) was obtained from the Cantharellus cibarius by ultrasonic wave extraction. Molecular modificate through concentrated sulfuric acid method, then sulfating Cantharellus cibarius polysaccharide (S-CCP) was produced. Scavenging effects of S-CCP on superoxide radical and hydroxyl free radical were studied, and the protective effects on H2O2 induced hemolysis of mice erythrocytes and the lipid peroxidation of liver homogenate in mice were also investigated. The results showed that S-CCP has the effective antioxidant activity in vitro. When the concentration was 5 mg/mL, S-CCP clearance rate of O2 higher22.71% than CCP. When the concentration was 0.4 mg/mL, S-CCP clearance rate of OH higher 14.9% than CCP. When the concentration was 4 mg/mL, S-CCP against MDA induced by H2O2 lower 0.4 nmol/mg than CCP. When the concentration was 4 mg/mL, S-CCP inhibitory rate of the hemolysis of rat RBC induced by H2O2 higher 33.9% than CCP.
