毕赤酵母液态发酵产木聚糖酶条件研究

摘    要:用麦麸、米糠农业废弃物作为主要原料生产木聚糖酶,对毕赤酵母液态发酵产木聚糖酶的条件进行研究。结果表明,最适产酶条件:以质量浓度20%的麦麸作为碳源,4%酵母浸膏作为氮源,调节初始pH值5.5,培养温度30℃,摇瓶发酵装液量6%,培养时间130 h,在此条件下毕赤酵母液态发酵产木聚糖酶最高活力达到2192 IU/mL。

Abstract:Agricultural wastes,such as wheat bran and rice bran,were used as the main raw material for the production of xylanase,and the conditions of xylanase production by Pichia pastoris through liquid fermentation were optimized.The results showed that the optimum conditions of enzyme production were:wheat bran as carbon resource with the amount of 20%(w/v);yeast extract as nitrogen resource with the amount of 4%(w/v),C/N ratio 5:1;initial pH 5.5;culture temperature 30 ℃ and incubation time 130 h.Under such conditions,liquid fermentation Pichia pastoris xylanase enzyme activity reached the highest 2192IU/mL.
