摘    要:以一株新的嗜热拟青霉(Paecilomyces thermophila)J18为出发菌株,对其液态发酵产木聚糖酶的条件进行了优化。结果表明,以玉米芯为碳源能高效诱导木聚糖酶的胞外分泌,胰蛋白胨为最佳氮源,初始pH和培养温度分别为pH 7.5,50℃,所得酶活力最高。在优化后的培养基和培养条件下,第5 d酶活力达到峰值,为1 276 U/mL,表明该拟青霉能够利用农业废弃物高效生产木聚糖酶。该酶水解桦木木聚糖主要生成木二糖和木三糖,适合生产低聚木糖。

Abstract:Fermentation conditions of a newly isolated Paecilomyces thermophila J18 for xylanase production were investigated in this study.Corncob was found to be the best inducer and carbon source,while tryptone was the best nitrogen source for the xylanase production.The optimal initial pH and temperature of the cultures were pH 7.5 and 50℃,respectively.Highest xylanase activity of 1 276 U/mL was achieved under the optimal condition on the fifth day of incubation,showing the feasibility of xylanase production using corncob as carbon source. The major products of birchwood xylan hydrolyzed by the xylanase were xylobiose and xylotriose,suggesting the xylanase is suitable for xylooligosaccharides production.
