芽孢杆菌M-21产β-甘露聚糖酶发酵条件研究

摘    要:从土壤中分离筛选出产β-甘露聚糖酶的芽孢杆菌(Bacillus sp.)M-21,通过单因素实验和正交优化实验,确定了其最佳发酵产酶条件。菌株的产酶最适培养基组成包括(g/L)碳源:瓜尔豆胶4,复合氮源:豆粉20、(NH4)2HPO45,其他无机盐组分:K2HPO4.3 H2O1、MgSO4.7 H2O 0.5、NaCl 0.5、CaCl20.1、FeSO4.7 H2O0.001。产酶最适培养条件:培养基初始pH8.0,接种量4%,装液量50 mL/250 mL三角瓶,32℃180 r/min振荡培养36 h。此条件下酶活力最高可达1 487 U/mL。

Abstract:Bacillus sp.M-21 with high activity of β-mannanase was isolated from soil samples.The fermentation conditions were optimized in order to improve the production of the enzyme.The optimal liquid medium consisted of 4 g/L guar gum was used as carbon source,20 g/L soybean powder and 5 g/L(NH4)2HPO4 as nitrogen source and other inorganic salts(g/L): K2HPO4·3H2O 1,MgSO4·7H2O 0.5,NaCl 0.5,CaCl2 0.1,FeSO4·7H2O 0.001.The optimal culture conditions were: initial pH 8.0,inoculation volume 4%,medium volume 50 mL in 250 mL flask,agitation speed 180 r/min.After incubation at 32℃ for 36h,the enzyme reached its highest activity of 1 487 U/mL.
