聚唾液酸分离纯化过程中脱除蛋白质工艺的研究

摘    要:应用不可逆变性后的蛋白溶解力与聚唾液酸溶解力的显著差异来脱除聚唾液酸发酵液中蛋白质。通过实验优化了乙醇分步沉淀法和乙醇沉淀-过滤法脱除蛋白质的工艺操作参数。得到有效脱除聚唾液酸中蛋白质的工艺条件为:在经离心去除菌体的上清液中加入乙醇至体积百分数为75%,室温下放置沉降后,除去上层清液,加入15g/L硅藻土过滤,加去离子水使滤饼中聚唾液酸溶解,再次过滤,收集滤液。得到聚唾液酸回收率为94·5%,蛋白质的去除率达到89·5%。

Abstract:Based on the significant difference of solvency between polysialic acid and irreversibly denatured protein, two processes removing protein from the polysialic acid broth were carried out. The optimal conditions of protein removing process were gained, i.e. Ethanol was added to 75% in supernatant of polysialic acid broth. 15 g/L kieselguhr filter aid was added and filtrated after deposition was formed and the supernatant was discarded. De-ion water was added to dissolve the polysialic acid in the filter cake. The polisialic acid solution was obtained after filtration. The yield of polysiac acid is 94.5%, and the rate of removing protein reaches 89.5 %.
