枯草芽孢杆菌发酵生产利巴韦林的变温控制策略研究

摘    要:以枯草芽孢杆菌(Bacillus substilis)TM903为供试菌株,考察了不同的培养温度(34~40℃)对利巴韦林摇瓶发酵的影响。结果发现,38℃条件下菌体生长最好,利巴韦林产量最高,可达4.12g/L。基于上述结果,研究了3种变温控制模式条件下利巴韦林摇瓶发酵过程。得出如下结论:0~24 h发酵温度为38℃,24 h后每6 h提高2℃,并采用这种温度控制方式进行5L自控发酵罐试验,60h后可积累利巴韦林5.86 g/L,比同类研究的产量(4.69 g/L)提高了24.95%。

Abstract:In this paper,ribavirin production by Bacillus substilis TMg03 at various temperatures was in- vestigated to study the effect of temperature on ribavirin fermentation.Different cultivation temperatures ranging from 34℃to 40℃were evaluated in shake flasks.The result indicated that 38℃was the best for cell growth and the maximum ribavirin production(4.12g/L)was obtained.Based on the results,three tempera- ture-shifted controlling manners were studied.The conclusion was as follows:0~24 h,culture tempera- ture is 38℃,and it is increased 2℃every 6h after 24h.In 5L jar fermentor,5.86g/L ribavirin was accumula- ted in 60h using this controlling manner,which was increased 24.95% compared with reference 5.
