山茶油脱臭馏出物中角鲨烯的分离纯化及对猪油抗氧化作用的研究

摘    要:通过单因素分析和正交实验,得到从山茶油脱臭馏出物中富集角鲨烯的最佳皂化条件,并通过硅胶柱对富集的角鲨烯进行二次纯化,同时,采用Rancimet氧化分析仪研究纯化后角鲨烯的抗氧化性。结果表明:富集角鲨烯的最佳条件是碱浓度1.3 mol/L,料液比1∶6,反应时间60 min,反应温度90℃,在此条件下,角鲨烯质量分数为35.22%,提取率为81.97%;硅胶二次纯化后收率为94.5%,纯度为90%。猪油中添加0.02%的角鲨烯纯化物有显著的抗氧化效果(P0.05),但在高浓度下有促氧化作用。此外,角鲨烯纯化物与没食子酸丙酯(PG)复配有增效作用(P0.05)。

Isolation and Purification of Squalene from Camellia Oil Deodorized Distillate and Its Antioxidant Activity in Lard
Abstract:In the study, single factor analysis and orthogonal test were designed to optimize the saponification process of enriching squalene from the deodorized distillate in Camellia oil. In addition, silica gel column was utilized for further purification of squalene. Finally, we also investigated the oxidation resistance of refined squalene by Rancimet oxidation analyzer. The results showed that the optimum conditions were KOH concentration 1.3 mol/L, ratio of material to liquid 1:6, reaction time 60 min, reaction temperature 90 °C. Under this condition, the yield and extraction rate of squalene were 35.22% and 81.77%, respectively. The yield and purity of squalene could be enhanced to 94.5% and 90% separately after secondary purification by silica gel. 0.02% concentration of refined squalene in lard oil had significant antioxidant effect (p < 0.05), while adverse phenomenon was observed at high addition of squalene. Importantly, we also found that purified squalene had a significant synergistic effect with propyl gallate (PG) (p < 0.05).
