小麦醇溶蛋白和谷蛋白亚基的高效毛细管电泳分离研究

摘    要:初步探讨了小麦醇溶蛋白和低分子量谷蛋白亚基(LMW-GS)高效毛细管电泳(HPCE)分离的分辨芳及其重复性。结果显示:利用碱性硼酸钠缓冲液系统及一个改进的毛细管冲洗程序分离麦醇溶蛋白,可获得较高的分辨度和重要性。通常在30min内一个品种较易分离出40个左右醇溶蛋白组分,而且重复分离之间峰迁移时间和峰高的平均相对标准偏差(RSD)可分别保持在0.2%和4%左右。利用pH2.5酸性磷酸缓冲液(含20

Abstract:The resolution and reproducibility of wheat gliadin and low-molecular weight glutenin subunit (LMW-GS) separations by high-performance capillary electrophoresis (HPCE) were studied. The results showed that,higher resolution and reproducibility of gliadin HPCE separations were obtained by using the alkaline sodium borate buffer system and an improved capillary cleaning procedure.In general,about 40 gliadin components for each cultivar can be revealed by above HPCE methods. At the same time, the average relative standard deviations (RSD) of migration times and heights of gliadin peaks were about 0.2% and 4%,respectively. By using the pH 2.5 phosphate buffer containing 20% acetonitrile and 0.05% hydroxypropylmethylcelluose,about 20 LMW glutenin subunits from each cultivar could be separated while higher reproduc ibility can be maintained.In comparison with the routine A-PAGE and SDS-PAGE methods, high-performance capillary electrophoresis has shown to be rapid,high-resolution,automatic, and easily quantifying. Thus, HPCE has much potential for cereal variental identification and quality prediction.
