麦苗中超氧化物歧化酶(SOD)抗氧化活性研究

摘    要:本研究探讨麦苗中超氧化物歧化酶(Superoxide Dismutase,SOD)在动物体内和体外的抗氧化活性。体外抗氧化试验中将麦苗中经过提取、纯化、干燥制成的SOD成品经蒸馏水配制成溶液,分别采用DPPH·清除法、Fenton反应法和Fe~(3+)还原法测定其对DPPH和羟自由基的清除率及其还原能力。通过饲养昆明种小鼠,经口灌胃受试物SOD酶液以及生理盐水、番茄红素,考察丙二醛(MDA)含量、过氧化氢酶(Catalase,CAT)活力和谷胱甘肽过氧化物酶(GSH-Px)活力。结果表明,体外试验中SOD酶液具有良好的抗氧化能力,SOD(1 mg/mL)酶液在1 mL水平下对DPPH、羟基自由基的清除率分别可达到96.95%、98%,还原能力随剂量的增加呈递增趋势;体内试验中SOD剂量组与对照组相比,小鼠肝组织中MDA差异显著(P0.05),且各个剂量组下MDA含量都有减少。与对照组比较,SOD高剂量组能提高小鼠肝组织中GSH-Px和CAT活力(P0.05)。麦苗中SOD在体外和体内均表现出良好的抗氧化作用。

Antioxidant activity of superoxide dismutase from wheat leaves
Abstract:In present study, the antioxidant activity of superoxide dismutase (SOD) from wheat leaves was verified with the animal in vivo and in vitro methods. DPPH? and hydroxyl free radical scavenging capacity and the reduction capability of SOD solution from young wheat leaves were determined by means of DPPH assay, Fenton reaction and Fe3 reduction method with in vitro experiment respectively. And in vivo experiment were promoted as following that a total of fifty healthy Kunming mice were randomly divided according feeding their solvent different into dose group and control group. The dose group and control group were intragastrically administered with SOD tested samples and equivalent solvent respectively. The capability of hydrogen peroxide enzyme (CAT) and glutathione peroxidase (GSH-Px) and malondialdehyde (MDA) content in liver tissue were measured. The results indicated that SOD enzyme fluid has good antioxidant removal ability through in vitro experiments, scavenging capacity can reach 96.95% and 98% in SOD (1mg.mL-1) enzyme fluid in 1mL level of DPPH? and hydroxyl free radical, respectively. Reducing capacity shows an increasing trend. Compared SOD dose group with control group in vivo tests , the significant difference (P<0.05) MDA concentration was determined in mice liver tissue, and the content of every dose group formanced reducing trend. Higher SOD dose could improve GSH-Px vitality and CAT (P<0.05) activity in mice liver tissue. Therefore, obvious antioxidation activity was observed on superoxide dismutase from younger wheat leaves through in vitro and in vivo methods.
