摘    要:目的评估测定瑞替普酶溶栓生物活性的4种方法。方法分别使用凝块溶解法、平板溶圈法、发色底物法及气泡法4种方法,测定同一批瑞替普酶样品的生物活性。结果4种测活方法的误差分别为10%,3%,3%和6%,表明测定瑞替普酶体外生物活性的4种方法均有可比性。结论平板溶圈法和发色底物法测活的重复性较好,特异性好,灵敏度高,较适合测定瑞替普酶活性的要求。

Abstract:Objective To evaluate the four assay methods for reteplase activity. Methods The bioactivity of the same reteplase sample was assayed by four methods of clot lysis assay, fibrin plate assay, chromogenic substrate assay and bubble assay, respectively. Results The errors from the four assay methods were 10%, 3%, 3%, 6%, respectively. It indicated that there existed relativity among the four assay methods for reteplase activity in vitro. Conclusion Both fibrin plate assay and chromogenic substrate assay most adapt to reteplase activity analysis due to good repetitiveness, specificity and high sensitivity.
