穿山龙多糖的提取及其体外抗氧化活性研究

摘    要:目的提取穿山龙多糖,对主要影响因素进行考察,优化提取工艺,并对所得多糖的体外抗氧化活性进行研究。方法以多糖收率和含量为综合评价指标,考察料液比、水浴温度和提取时间对水醇法提取穿山龙多糖的影响,通过正交试验确定最优工艺。采用超氧阴离子自由基(O-2?)、铁离子和DPPH?研究其体外抗氧化活性。结果水醇法提取穿山龙多糖最佳工艺为料液比1:10,水浴温度90℃,提取时间4 h,多糖提取率为2.72%。所得穿山龙多糖对O-2?及DPPH?表现良好的清除能力,对铁离子也具有较好的还原力。结论建立了一种简便有效的穿山龙多糖提取工艺。所得穿山龙多糖具有良好的体外抗氧化能力,且呈一定的剂量依赖性。

Abstract:Objective To optimize the extraction technology of polysaccharide from Dioscorea nipponica Makino and investigate the main factors affecting the extraction and the antioxidant activity of the polysaccharide.Methods The effects of the solid-liquid ratio,temperature of water bath and extaction time on the extraction of polysaccharide by hydroalcoholic method were investigated through orthogonal test.The yield and content of polysaccharides were taken as comprehensive evaluation indexes.In addition,the antioxidant activity was detected by usingO-2?,iron ions and DPPH?.Results The optimum hydroalcoholic extraction process for of polysaccharide from Dioscorea nipponica Makino was as follows:the bath temperature was 90℃,the extraction time was 4 h,and the ratio of solid to liquid was 1:10.Under these conditions,the yield of polysaccharide was 2.72%.The polysaccharide obtained showed goodO-2?and DPPH?scavenging capacities as well as good reductive activity on Fe3+.Conclusion A simple and effective extraction technology of polysaccharide from Dioscorea nipponica Makino was established.The polysaccharide had a good antioxidant activity in vitro in a concentration dependent manner.
