刺参中过氧化物还原酶peroxiredoxin4的抗氧化活性分析

摘    要:目的 研究刺参中的抗氧化活性物质Peroxiredoxin4。方法 利用生物信息学方法对刺参Peroxiredoxin4一级结构进行分析,克隆并表达重组刺参Peroxiredoxin4蛋白,构建DTT/Fe3+/O2混合功能氧化酶体系,确定重组刺参Peroxiredoxin4蛋白功能。结果 刺参Peroxiredoxin4氨基酸序列含有活性Cys,且活性Cys位于Prx4保守基序FYPLDFTFVCPTEI和HGEVCPAGW中,克隆得到的重组刺参Peroxiredoxin4蛋白能够保护DNA免遭氧化剂损伤。结论 重组刺参Peroxiredoxin4蛋白具有抗氧化生物学活性。

Abstract:Objective To investigate the antioxidative activity of peroxiredoxin4 in Apostichopus japonicus. Methods Bioinformatics methods were used to analyze the amino acid composition and tertiary structure of Apostichopus japonicus. The recombinant protein was cloned and expressed, the DTT/Fe3+/O2 system was constructed, and the function of recombinant peroxiredoxin4 protein was determined. Results There were Cys residues with antioxidative activity in the amino acid sequence of Apostichopus japonicus, and the Cys residues were in the conserved sequence FYPLDFTFVCPTEI and HGEVCPAGW, respectively. The recombinant peroxiredoxin4 protein protected DNA against oxidant damage. Conclusion The recombinant peroxiredoxin4 of Apostichopus japonicus had the antioxidative activity.
