摘    要:目的通过响应面试验设计,优化高压脉冲电场辅助提取火棘果主要活性成分的工艺。方法根据Box-Behnken中心组合试验设计原理,在单因素试验的基础上,选取料液比、乙醇浓度、电场强度和脉冲数为考察因素,采用综合评分法进行4因素3水平的响应面试验。结果根据回归方程优化并得到最佳提取工艺为料液比1:30(g/mL)、乙醇浓度68%、电场强度19 kV/cm、脉冲数11个,在此条件下综合评分达96.82分,与回归方程得出的理论预测数值97.96基本相符。结论响应面-综合评价法优化的高压脉冲电场辅助提取工艺,应用在火棘果主要活性成分的提取上简单可靠,为湖南地区火棘果资源的综合开发利用提供了理论和依据。

Abstract:Objective To optimize the extraction process of main active components from Pyracantha fortuneana fruit by high voltage pulsed electric field, through response surface experimental design. Methods According to the design principle of Box-Behnken central composite test and the single factor test, the solid to liquid ratio, ethanol concentration, electric field strength and pulse number were selected as the investigation factors, and the response surface test with 4 factors and 3 levels was conducted using the comprehensive scoring method. Results According to the regression equation, the optimal extraction conditions were obtained as follows: Solid to liquid ratio was 1:30 (g/mL), ethanol concentration was 68%, electric field strength was 19 kV/cm, and number of pulses was 11. Under these conditions, the comprehensive score was 96.82, which was basically consistent with the theoretical prediction value of 97.96 by the regression equation. Conclusion The high voltage pulsed electric field assisted extraction process optimized by response surface methodology is simple and reliable in the extraction of main active components of Pyracantha fortuneana fruit, which provides a theoretical basis for the comprehensive development and utilization of Pyracantha fortuneana fruit resources in Hunan.
