摘    要:目的研究生姜中主要活性成分姜酚的分离方法。方法选用D4020作为固定相,乙醇水溶液作为流动相,从溶剂萃取法得到的姜油树脂中分离姜酚。采用紫外分光光度法(ultraviolet,UV)与高效液相色谱法(high performance liquid chromatography,HPLC)检测分离纯化过程,讨论柱高、流速、上样量对姜酚分离效果的影响,确定最佳工艺条件。结果柱层析法分离姜酚的实验条件:柱高为25 cm,洗脱速度1 mL/min,上样量100 mL。两次纯化后姜酚纯度可以达到85.24%。结论该方法可以提高姜酚的纯度。

Abstract:Objective To investigate the separation method of gingerol in ginger. Methods D4020 macroporous resin was used as the stationary phase and ethanol-water solution was used as mobile phase. Ultraviolet (UV) and high performance liquid chromatography (HPLC) were used to trace the seperation process and explore the influence elements of gingerol seperation. Effects of column height, flow rate and sample amount on separation of gingerol were discussed, and optimum technological conditions were determined. Results The experimental conditions of the separation of gingerol by column chromatography were as follows: the column height was 25 cm, the flow rate was 1 mL/min, and the sample amount was 100 mL. After two times of purification, the purity of gingerol could reach 85.24%. Conclusion The established method can improve the purity of gingerol.
