摘    要:目的评价亚甲蓝光化学法(MBP)对血浆中水疱口膜炎病毒(VSV)的灭活效果。方法将7.12 logTCID50.mL-1VSV加入血浆中,采用不同的参数组合[亚甲蓝(MB)浓度(0.8、1.3μmol.L-1)、光照强度(20 000、50 000 LUX)、光照时间(10、20、30 min)]灭活处理后,分别将样品加入非洲绿猴肾细胞(Vero)单层细胞中培养,观察细胞病变(CPE)状况,评价病毒灭活的效果。结果经过30 min灭活处理,可将病毒彻底灭活,病毒滴度下降6 log值以上。实验设置的MB浓度和光照强度对血浆病毒灭活的影响不明显。结论在MB浓度为0.8μmol.L-1和光照强度20 000 LUX的条件下,处理血浆30 min以上就能有效灭活血浆中的VSV病毒。

Abstract:Objective To evaluate the photochemical effects of methylene blue(MBP) on inactivation of vesicular stomatitis viruses(VSV)in plasma.Methods Plasma containing 7.12 logTCID50 ·mL-1 VSV and different concentrations of methylene blue(MB)(0.8 and 1.3 μmol·L-1)was illuminated by fluorescent light(20 000 and 50 000 LUX)for 10,20 and 30 minutes.Then,the titers of VSV in plasma were detected by infecting Vero cells and cytopathic effect(CPE) was observed for evaluation of viral inactivation.Results VSV in plasma were completely inactivated after treatment for 30 minutes,and the titers decreased by greater than 6 log.The effect of viral inactivation was not significantly correlated with MB concentrations and light intensity.Conclusion Under the condition of 0.8 μmol·L-1 MB,20 000 LUX of light intensity and 30 minutes of treatment,VSV in plasma can be effectually inactivated.
